General description
Eukaryotic peptide chain release factor GTP-binding subunit ERF3A (UniProt: P15170; also known as Eukaryotic peptide chain release factor subunit 3a, eRF3a, G1 to S phase transition protein 1 homolog) is encoded by the GSPT1 (also known as ERF3A) gene (Gene ID: 2935) in human. GSPT1 is a ubiquitously distributed protein that serves as a translation termination factor. It is shown to be essential for cell survival and it interacts with multiple factors involved in key cellular processes including regulation of cell cycle progression. It is a GTPase component of the eRF1-eRF3-GTP ternary complex that mediates translation termination in response to the termination codons UAA, UAG and UGA. This ternary complex binds to a stop codon in the ribosomal A-site. GTP hydrolysis by GSPT1 is reported to induce a conformational change that leads to its dissociation, permitting eRF1 to accommodate fully in the A-site. Degradation of GSPT1 results in TP53-independent cell death in leukemia cells while sparing normal hematopoietic stem cells. Three isoforms of GSPT1 have been described that are produced by alternative splicing. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GSPT1 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GSPT1 in A549 and Hek293T cell lysates.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected GSPT1 in HeLa cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GSPT1 in Human colon tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1F21 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Eukaryotic peptide chain release factor GTP-binding subunit ERF3A (GSPT1). It targets an epitope within 15 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
