General description
Native, bovine thrombin. Key enzyme that activates factor XIII and converts fibronogen into fibrin. Serine protease formed in the plasma during the coagulation process by activation of prothrombin by a complex of Factor Xa, Factor Va, phospholipid, and Ca2+. Thrombin cleaves Arg-Gly bonds in specific sequences only. The best characterized activity of thrombin is the conversion of fibrinogen to fibrin in the clotting process. Inhibited by AEBSF (Cat. No. 101500) Antithrombin (Cat. No. 169756), PMSF (Cat. No. 52332), and TLCK (Cat. No. 616382). Note: 1 KU = 1000 units.
Physical form
Lyophilized from 200 mM NaCl, 50 mM sodium citrate, 0.1% PEG-8000, pH 6.5.
Preparation Note
Following reconstitution, aliquot and freeze (-70°C). Stock solutions are stable for up to 6 months at -70°C.
Analysis Note
Complete activation from homogeneous bovine prothrombin by SDS-PAGE
Other Notes
One unit is determined by comparison with a standard curve prepared using the Bureau of Biologics standard thrombin.
