CCT374705 is an orally available, potent and selective BCL6 inhibitor (IC50 = 6 nM/TR-FRET, 22 nM/NanoBRET) that inhibits BCL6-dependent proliferation in cultures (GI50 = 38.5 nM/OCI-Ly1 & 14.9 nM/Karpas 422). CCT374705 oral administration reduces Karpas 422 xenograft-derived tumor growth in mice in vivo (by 46% on day 35; 50 mg/kg b.i.d. p.o.).
