General description
Transcription intermediary factor 1-beta (UniProt Q13263; also known as EC:2.3.2.27, E3 SUMO-protein ligase TRIM28, KAP-1, KRAB-associated protein 1, KRAB-interacting protein 1, KRIP-1, RING finger protein 96, TIF1-beta, Transcriptional intermediary factor 1-beta, Tripartite motif-containing 28) is encoded by the TRIM28 (also known as KAP1, PPP1R157, RNF96, TF1B, TIF1B) gene (Gene ID: 10155) in human. TRIM28/KAP-1 is a widely expressed, oligomeric, nuclear corepressor for KRAB domain-containing zinc finger protein that mediates gene silencing by recruiting chromodomain helicase DNA binding protein 3 (CHD3) complex and SET domain bifurcated histone lysine methyltransferase 1 (SETDB1) to the promoter regions of KRAB target genes. It enhances transcriptional repression by coordinating the increase in H3K9me, the decrease in histone H3 lysine 9 and lysine 14 acetylation, and the disposition of HP1 proteins to silence gene expression. Application
Quality Control Testing
Evaluated by Western Blotting in RAW264.7 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TRIM28/KAP-1 in RAW264.7 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TRIM28/KAP-1 in U2OS cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TRIM28/KAP-1 in Human small intestine tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TRIM28/KAP-1 in K562 cells.
Affinity Binding Assay: A representative lot of this antibody bound TRIM28/KAP-1 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1G5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects TRIM28/KAP-1. It targets an epitope within 25 amino acids from the N-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
