BAY-390 is an orally active, potent and selective TRPA1 inhibitor (cellular Ca++ influx IC50 in nM by FLIPR = 16/hu, 19/monkey, 63/rat, 68/guinea pig, 73/mu, 81/dog; IC50 = 5.6 µM/hTRPC5, >25 µM/human TRPV1/4, TRPC3/6, KCNK9 (TASK-3), CACNA1H (Cav3.2)) that shows in vivo efficacy in animal models of inflammatory pain (30 or 90 mg/kg mice, p.o.) and neurogenic pain (30 or 90 mg/kg rats, b.i.d. p.o.).
