General description
一种三唑基-硫代吗啉基-甲酮化合物，通过直接亲和相互作用（KD = 7.12 µM）充当成纤维细胞生长因子诱导型14（Fn14）拮抗剂，并与肿瘤坏死因子样弱凋亡诱导物（TWEAK）C末端/细胞外TNF的同源域（THD）竞争Fn14结合（对500 pg/mL sTWEAK的抑制率为16.8%；[药物]=25 µM）。显示在10 µM浓度下可完全抑制100 ng/mL sTWEAK刺激的T98G神经胶质瘤细胞迁移，并且比TNFα诱导的NF-κB活化更有效抑制TWEAK-（在使用Fn14-NF-kB-Luc和NF-κB-Luc细胞的报告基因分析中，分别为IC50 = 7.8 vs. 31.03 µ M），在T98G培养物中高达50 µM和72 h没有任何明显的细胞毒性。
