General description
Tyrosine-protein kinase Tec (UniProt: P42680; also known as EC:2.7.10.2, Tec) is encoded by the TEC (also known as PSCTK4) gene (Gene ID: 7006) in human. The Tec family of protein kinases consists of five members, Tec, Btk, Itk/Emt/Tsk, Bmx and Txk/Rlk. They are abundantly expressed in hematopoietic cells and participate in the growth and differentiation of blood cells. Tec is a peripheral membrane protein that serves as a non-receptor tyrosine kinase that contributes to signaling from many receptors and participates as a signal transducer in multiple downstream pathways, including regulation of the actin cytoskeleton. It is activated by tyrosine phosphorylation by a wide range of cytokine stimulations. It regulates the development, function and differentiation of conventional T-cells and nonconventional NKT-cells and is required for TCR-dependent IL2 gene induction. Following B-cell or T-cell receptors activation by antigen, it is shown to translocate to the plasma membrane through its PH domain (aa 4-111). Tec is reported to serve as a central regulator of the noncanonical caspase-8 inflammasome that is involve in the assembly and activation of the caspase-8 inflammasome in response to fungal pathogens but is not involved in anti-bacterial immune responses. Immunogen
KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal region of human Tyrosine protein kinase Tec.
Application
Affinity Binding Assay: A representative lot of this antibody bound Tec peptide with a KD of 1.2 x 10-5 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Tec in K562 cells.
ELISA Analysis: A representative lot specifically detected Tec peptide in ELISA application, but not peptides from five other proteins.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions
Clone 19M23 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Tyrosine-protein kinase Tec. It targets an epitope within 19 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
