General description
Non-structural protein V (UniProt: P11207; also known as V5 tag) is encoded by the P/V gene in Parainfluenza virus 5. Epitope tags are genetically engineered short protein sequences that are serve as universal epitopes and can be easily detected with antibodies without affecting the native structure or function of proteins. The V5 tag is a sequence of 14 amino acids (GKPIPNPLLGLDST) derived from the internal region of non-structural protein V of parainfluenza virus 5. It can be used to detect expression of recombinant proteins in bacteria, insects, and mammalian cell systems. It is useful in detecting recombinant proteins that contain the V5 epitope. KLH-conjugated linear peptide corresponding to 14 amino acids from the internal region of V5 protein from the Parainfluenza virus 5.
Application
Quality Control Testing
Evaluated by Western Blotting with His/V5-tagged recombinant Tau-441 protein.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected His/V5-tagged recombinant Human Tau-441 protein.
Tested Applications
ELISA Analysis: A serial of dilutions from a representative lot detected V5-tagged recombinant Human Tau-441 protein.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HEK293 cells transfected with a V5 expression plasmid.
Peptide Inhibition Assay:: Target band detection in V5-tagged recombinant Human Tau-441 protein was prevented by preblocking of a representative lot with V5 tag peptide, but not the corresponding non-V5 tag control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1K20-L2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects proteins with V5 tag. It targets an epitope within 14 amino acids from the internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
