HSip-1 (Cyclen-AF-Cu; Hydrogen Sulfide imaging probe-1) is a sensitive hydrogen sulfide (H2S) fluorescent probe that displays a selective turn-on fluorescence response to sulfide anion (Ex/Em = 491/516 nm), but not GSH, L-Cys, DL-Hcy, 2-ME, DTT, SCN-, Na2SO3, Na2S2O3, or sodium ascorbate. HSip-1 is not membrane-permeable and is commonly employed for detecting H2S in media/buffers, cell lysates, blood, physiological fluids and tissue homogenates. HSip-1 is commonly used in the range from 1-100 μM depending on the H2S concentrations in samples, optimal probe concentration needs to be titred.
