General description
Glutaminyl-peptide cyclotransferase-like protein (UniProt: Q9NXS2; also known as EC:2.3.2.5, Golgi-resident glutaminyl-peptide cyclotransferase, isoQC, gQC) is encoded by the QPCTL gene (Gene ID: 54814) in human. QPCTL is single-pass type I membrane protein that is exclusively localized within the Golgi complex. It contains a conserved zinc-binding motif (Asp-Glu-His), and its transmembrane domain is localized in amino acids 35-55. Its Golgi retention signal lies between amino acids 19 to 53. QPCTL is responsible for the biosynthesis of pryoglutamyl peptides. It catalyzes the cyclization of N-terminal glutamine and glutamic acid residues into an N-terminal pyroglutamate residue (pGlu). It serves as a modulator of CD47-SIRPα binding. Inhibition of QPCTL is shown to enhance myeloid cell-mediated and antibody-dependent antitumor responses. It is also reported to regulate macrophage and monocyte abundance and inflammatory signatures in the tumor microenvironment. Inhibitors of QPCTL are reported to reduce pGlu-CD47 formation without affecting surface levels of CD47. Inhibition of pGlu formation is known to enhance the antitumor activity of myeloid cells similarly to antibody blockade of CD47 or SIRPα. Deletion of QPCTL is shown to control in vivo tumor growth. Application
Quality Control Testing
Evaluated by Western Blotting in Human liver microsomal preparations.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected QPCTL in Human liver microsomal preparations.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound QPCTL peptide with a KD of 5.8 x 10-7 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected QPCTL in Human stomach tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected QPCTL in HepG2 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 5M19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Glutaminyl-peptide cyclotransferase-like protein (QCPTL). It targets an epitope within 15 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
