Biochem/physiol Actions
PRGL493, a highly effective and selective Acyl-CoA synthetase 4 (ACSL4) inhibitor, exhibits potent anti-tumor activities by inhibiting cell proliferation and tumor growth in breast and prostate cancer models, both in vitro (IC50 ~ 25 µM) and in vivo (250 μg/kg, i.p., in mice). PRGL493 functions mainly by reducing ABCG2 (ATP-binding cassette subfamily G member 2) in TNBC and promoting ER expression. PRGL-493 effectively disrupts steroidogenesis, demonstrated by the significant reduction of steroid levels in cell lines and mouse models. Additionally, PRGL493 enhances the sensitivity of cancer cells to conventional chemotherapeutic and hormonal treatments.
