General description
14-3-3 protein β/a (UniProt: P31946; also known as Protein 1054, Protein kinase C inhibitor protein 1, KCIP-1) is encoded by the YWHAB gene (Gene ID: 7529) in human. Mammals express seven distinct 14-3-3 isoforms (gamma, epsilon, beta, zeta, sigma, theta, tau) that form multiple homo- and heterodimers, thus rendering 14-3-3 as a key multifunctional regulatory molecule. 14-3-3 protein beta/alpha serves as an adapter protein that has been implicated in the regulation of a large spectrum of both general and specialized signaling pathways. It binds to many partners, usually by recognition of a phosphoserine or phosphothreonine motif and its binding generally results in the modulation of the activity of the binding partner. The alpha, brain-specific form, differs from the beta form in its ability to be phosphorylated on serine 60 by protein kinase C delta in a sphingosine-dependent fashion. 14-3-3 β/a is shown to block the nuclear translocation of the phosphorylated form of SR protein-specific kinase 2 (SRPK2) and antagonize its stimulatory effect on cyclin D1 expression that results in blockage of SRPK2 elicited neuronal apoptosis. It also reported to serve as a negative regulator of osteogenesis. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected 14-3-3 β/a in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected 14-3-3 β/a in SHSY5Y and COS-1 cell lysates.
ELISA Analysis: 1.0 μg/mL from a representative lot detected recombinant 14-3-3 β/a.
Affinity Binding Assay: A representative lot of this antibody bound 14-3-3 b/a peptide with a KD of 2.3 x 10-6 in an affinity binding assay.

Biochem/physiol Actions
Clone 1D15 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects 14-3-3 protein β/a. It targets an epitope within 19 amino acids from the C-terminal half.
Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
