High-affinity, human-selective SUCNR1 (GPR91) antagonist that blocks human GPR91-mediated cellular signaling with no activity towards rat species.
NF-56-EJ40 is a high-affinity, potent and human species-selective succinate receptor SUCNR1 (GPR91) antagonist (human GPR91 Ki/KD = 17.4/33 nM; GTPγS assay IC50 = 25 nM/human, >100 μM/rat). Extracellular succinate is reported to hyperpolarize M2 macrophages through GPR91-mediated Gq signaling, blockable by NF-56-EJ40 (1 μM) or Gq inhibitor YM-254890 (1 μM).
