Stimulate T cells reactive to the TB7.7 antigen of Mycobacterium tuberculosis using PepTivator® Peptide Pools. PepTivators are pools of lyophilized peptides, consisting mainly of 15-mer sequences with 11 amino acids overlap. PepTivator M. tuberculosis TB7.7 is a peptide pool covering the complete sequence of the TB7.7 protein (UniProtKB Acc. no. P9WJ11).
In vitro stimulation of antigen-specific T cells with PepTivator Peptide Pools causes the secretion of effector cytokines and the up-regulation of activation markers, which then allow the detection and isolation of antigen-specific T cells.
Tuberculosis (TB) caused by infection with Mycobacterium tuberculosis is one of the leading causes of death worldwide. According to the WHO a quarter of the world’s population is infected with M. tuberculosis. While most infections are latent and asymptomatic, about 5–10% of infections progress to active disease. TB persists as a serious global health threat, requiring new vaccines and therapeutics to combat multidrug resistance. TB7.7 (Rv2654c) is an immunodominant M. tuberculosis antigen that potently induces Th1 immune responses and concomitant IFN-γ secretion by T cells. Along with the M. tuberculosis antigens ESAT-6 and CFP-10, TB7.7 is used in commercially available IFN-γ release assays (IGRAs) to detect latent TB infection.
