Clone 7E.17G9 reacts with CD278, the inducible co-stimulatory molecule (ICOS), a 47–57 kDa homodimeric glycoprotein of the CD28 family. Directly ex vivo, CD278 is expressed only by a small subset of T cells, including follicular B helper T cells (TFH) and regulatory T cells. However, CD278 is strongly up-regulated on the surface following T cell activation. Upon binding to its ligand (ICOSL), CD278 initiates a multitude of events, which modulate many different aspects of T cell–mediated immune responses. Besides regulating differentiation and activation of effector as well as regulatory CD4+ T cells, ICOS also plays a role in germinal center formation and isotype switching.
